Squalen publishes original and innovative research to provide readers with the latest research, knowledge, emerging technologies, postharvest, processing and preservation, food safety and environment, biotechnology and bio-discovery of marine and fisheries. The key focus of the research should be on marine and fishery and the manuscript should include a fundamental discussion of the research findings and their significance. Manuscripts that simply report data without providing a detailed interpretation of the results are unlikely to be accepted for publication in the journal.

Abstract

A comprehensive study on exposure assessment of the priority organic contaminants via seafood consumption has been conducted to the coastal population of Jakarta Bay. Seafood is essential food source in Indonesia and also important income for the majority of coastal populations. A number of 152 respondents from 4 districts surounding the bay were interviewed to record their frequency and pattern on seafood consumption. In the same time, 13 seafood species were collected directly from Jakarta Bay during the dry and wet seasons for the assessment of organic contaminants. A non-target GC/MS screening identified more than 40 organic contaminants in which 6 of them are potentially considered as priority contaminants including 3 groups of carcinogenic contaminants i.e. dichlorodiphenyl-trichlorethane (DDT) and its metabolites (DDXs), dichlorobenzenes (DCB) and carcinogenic PAHs (PAH4). Further exposure analysis suggested cumulative health risk of these contaminants was less than official minimal risk level (MRL) and therefore categorized safe for the corresponding population. However, attention must be paid since additional exposure of either from the different food category or other exposure route may contribute to significantly elevate the health risk on the population as well as potential exposure of emerging contaminants.

Abstract

The purpose of this study is to study the prevalence of aflatoxin B1 in commercial dried salted fish and other related information. A total of 150 samples were classified into 3 groups, based on the salt content. Low salt content (0-5%) were dried anchovies (Stolephorus sp.) and commerson’s anchovy (Stolephorus commersonii); moderate salt content (6-10%) were medan anchovy (Stolephorus bataviensis) and  whipfin silverbiddy (Gerres filamentosus) while high salt content (10%) were moonlight gouramy (Trichogaster microlepis) and snakehead fish (Channa striata). The samples were collected from different seller in Java Island and then determined for Aspergillus flavus, Aflatoxin B1, salt content, moisture content, pH, water activity and total mold count. Results showed that dried salted fish were contaminated with A. flavus at temperature of 25.2-32.2 oC, 65-84% humidity, 17-50% moisture content, 0.25-19.88% salt content, and 0.73-0.86 aw. The prevalence of A. flavus in dried salted fish was 9.33% (14/150) and the prevalence of aflatoxin B1 was 8% (12/150) with detectable concentrations of 10.71-33.6 ppb.

Abstract

Since algae are well known as the host of fungi, the information about total fungal community associated with alga is needed to give the initial data that is useful for natural product discovery. The study on total fungal community associated with marine macroalga U. intestinalis from Xiamen-China was carried out using MiSeq, a powerful platform for microbial ecology studies. Metagenomics DNA was isolated from surfaces of the U. intestinalis thallus and rocks colonized by this alga and  18S ribosomal RNA region was sequenced using MiSeq Illumina to yield 81360 sequences from 162720 reads, which average read length is 245.61 bp. The sequences were assigned to operational taxonomic units (OTUs), where 101 genera represent 5 phyla, 17 classes and 64 families of fungal. Ascomycota was the highest abundance share of fungal community at phylum level. Meanwhile Aspergillus, Blastobotrys, Alternaria, Knufia, and Fusarium were some of dominant genera found associated with U. intestinalis which have been reported as the promising source of natural products. By using MiSeq platform, this study revealed the total community structure of algicolous fungi associated with U. intestinalis.

Abstract

The study aimed to investigate the chemical and organoleptic characteristics of smoked tilapia fish fillets which were produced using different drying process and enriched with olive oil. Smoked fish were made using the following method: the fish were fasted overnight, shocked with cold water (4-5 oC) and then slaughtered and filleted. The fillets were soaked for 30 minutes in a brine solution containing spices and 5% (v/v) of liquid smoke. 5% (v/v) of olive oil was added into the solution and compared with control without olive oil addition. After 30 minutes, fish were cleaned from the spices using water and smoked at a temperature of 80 oC for 4-5 hours. The drying process was conducted in two different methods, i.e using oven-drying and traditional smoking. The smoked tilapia products were analyzed for proximate composition and sensory profiles. The result showed that addition of olive oil and methods of drying affected the proximate composition as well as sensory characteristics of the tilapia fillets. The fillets with addition of 5% olive oil and processed with oven-drying were considered as the best treatment due to low moisture content but high ash, fat and protein contents with 45.6±0.4%; 6.8±0.1%; 7.9±0.1% and 82±1.6%,  respectively. Based on panelist evaluation, the fillets with 5% olive oil and processed with oven-drying also had high performances in appearance and texture.

Abstract

In many countries, most of household goods are made from polymer or plastic. The polymer manufacturing industry has been relying on fossil fuel-based raw materials which are non-biodegradable. Plastics derived from these non-biodegradable sources will be difficult to degrade in nature, which in turn will pollute the environment and harm the ecosystem. One way to reduce the ecosystem damages caused by petroleum-based plastic is by using biodegradable materials for plastic industry, such as Polyhydroxyalkanoates (PHA). Poly (3-hydroxybutyrate-co-3-hydroxyvalerate) or PHBV is an example of PHA that can be produced by several types of microorganism, one of them is marine archaea Haloferax mediterranei. The objective of this study is to determine the productivity of marine archaea H. mediterranei cultures to produce PHBV with yeast extract as main nutrient sources. Experiments and analysis were conducted in triplicate and batch operating system. During the fermentation process, optical density, glucose levels, phosphorus levels, pH, and dry cell weight (DCW) were observed. The final product was then analysed using GC-MS to determine 3HB (3-hydroxybutyrate) and 3HV (3-hydroxyvalerate) fraction composition in the PHBV. The results showed that the maximum specific growth rate (m) and doubling time (Td) of the H. mediterranei were 0.1258 hours-1 and 5.51 hours, respectively. H. mediterranei biomass increased until it reached stationary phase after 95 hours incubation. The PHBV content, dry biomass and yield of PHBV to substrate were 2.62 g/L, 29.1% and 0.27 g/g, respectively.
